AN ANIMAL MODEL FOR COMBINED METHOXYFLURANE AND GENTAMICIN NEPHROTOXICITY

Fischer 344 rats receiving gentamicin 20 mg/kg/day prior to and following 0.5% methoxyflurane anaesthesia for 3
hours had greater renal insufficiency than rats treated with either drug alone. Polyuria, progressively increasing BUN and decreasing urea nitrogen clearance were noted; a renal histological lesion with features of both drug treatments was seen with electron microscopy. Similar, atypically severe nephrotoxicity has been observed in patients treated with methoxyflurane and gentamicin, suggesting that toxic drug interaction also occurs in humans following combined administration of these drugs.
